Immunohistochemical assessment of possible anticancer effect mechanisms of 2-(6,8-dimethyl-5-nitro-4-chloroquinoline-2-yl)- 5,6,7-trichloro-1,3-tropolone in PDX models of lung cancer

Purpose of the study. Evaluation expression immunohistochemical tumor markers Ki-67, b-catenin, Bcl-2, P53, connexin 32 and 43 when using 2-(6,8-dimethyl-5-nitro-4-chloroquinoline-2-yl)-5,6,7-trichloro-1,3-tropolone in mice with xenographs squamous cell lung cancer. Materials methods . Subcutaneous PDX models human cancer were created immunodeficient BALB/c Nude mice. A fragment patient’s (3 × 3 mm) was implanted subcutaneously right thigh a previously anesthetized mouse. 200 μl administered orally probe 12 doses once every days. All animals divided into groups depending on tropolone doses: experimental 2–5 0.0055, 0.055, 0.55 2.75 mg/g, respectively. The control group received 1 % starch gel which carrier. euthanized 36 days after start substance administration, tissue isolated prepared for IHC study according to standard protocol. reactions performed antibodies 43. Results. Higher associated decreased Bcl-2 protein, but increased P53 protein. dosage directly proportional. Conclusion Immunohistochemical analysis proteins showed changes indicating its antitumor efficacy suggesting possible mechanism action based activation apoptosis.
